Capping structures at the 5'-terminus of polyadenylated ribonucleic Acid in Avena coleoptiles.

چکیده

Evidence is presented for the occurrence of 5'-terminal capping structures in the polyadenylated RNA of oat (Avena sativa) coleoptiles. These structures are composed of an inverted terminal nucleoside containing the modified base 7-methylguanine which is joined 5' to 5' with a second (penultimate) nucleoside by means of three phosphate groups in two pyrophosphate linkages. The penultimate nucleoside is joined to the remainder of the RNA molecule by a conventional 3',5' phosphodiester bond. A significant difference between the cap structures of oat coleoptile RNA and those of previously described higher eucaryotic cellular mRNAs is the lack of ribose methylations in the penultimate nucleosides of the plant RNA.

pH-Dependence of Extension Growth in Avena Coleoptiles and Its Implications for the Mechanism of Auxin Action.

The pH-dependence of acid-induced growth in excised segments of Avena sativa coleoptiles has been reinvestigated in the pH range 3 to 7. In contrast to previous reports (e.g. DL Rayle [1973] Planta 114: 63-73), only acidic buffers with a pH below 5.0 induce an extension response. A pH of 3.5 to 4.0 is required to mimic auxin-mediated growth. Very similar pH-response curves are obtained with bot...

Growth & geotropic curvature of Avena coleoptiles in presence of n-1-naphthylphthalamic acid.

The fact that geotropic curvature is mediated by unequal rates of elongation on opposite sides of the bending structure has led to the general implication that (lepression of these growth rates should cause a corresponding inhibition of geotropic curvature. Recently published results have revealed that this expected relationship was not always observed when plants were treated with compounds su...

Auxin-induced growth of Avena coleoptiles involves two mechanisms with different pH optima.

Although rapid auxin-induced growth of coleoptile sections can persist for at least 18 hours, acid-induced growth lasts for a much shorter period of time. Three theories have been proposed to explain this difference in persistence. To distinguish between these theories, the pH dependence for auxin-induced growth of oat (Avena sativa L.) coleoptiles has been determined early and late in the el...

Effects of diclofop and diclofop-methyl on the membrane potentials of wheat and oat coleoptiles.

Electrophysiological measurements were made on the mesophyll cells of wheat (Triticum aestivum L. cv Waldron) and oat (Avena sativa L. cv Garry) coleoptiles treated either with the herbicide diclofop-methyl (methyl 2-(4-(2',4'-dichlorophenoxy)phenoxy)propanoate), or it's primary metabolite diclofop, (2-(4-(2',4'-dichlorophenoxy)phenoxy)-propanoic acid). Application of a 100 micromolar solution ...

Immunogold electron microscopy of phytochrome in Avena: identification of intracellular sites responsible for phytochrome sequestering and enhanced pelletability

Using monoclonal antibodies to the plant photoreceptor, phytochrome, we have investigated by immunogold electron microscopy the rapid, red light-induced, intracellular redistribution (termed "sequestering") of phytochrome in dark-grown Avena coleoptiles. Pre-embedding immunolabeling of 5-micron-thick cryosections reveals that sequestered phytochrome is associated with numerous, discrete structu...
